British scientists from the University of Bata developed a first peptide inhibitor, which was not reversible with the key to the development of CJUN, a cancer protein -Transkripion factor. Transcription factors such as CJun regulate gene activity and become frepactic for cancer that usually causes uncontrolled cell growth. However, the development of drugs that can block these proteins has long been a difficult task. It turned out that traditional low molecular weight drugs were ineffective, which focused on scientists on peptides – small fragments of protein.
The CJUN protein consists of two identical creatures associated with DNA and changes the expression of genes. Scientists created a peptide associated with one of the CJun lines that prevented their interaction with DNA. This peptide was modified for irreversible attachment, which made it an effective “harpoon” that prevented CJUN forever.
“The inhibitor captures CJUN in a reliable way and does not allow it to contact DNA. This was used to prevent the transcription factor for the first time using a peptide inhibitor,” he said.
To search for effective peptides, scientists used an innovative platform called Survival (TBS), the transcription block. This method allows you to test thousands of peptides and controls the ability to block CJUN in real cellular conditions. If the Peptit successfully blocks CJUN, the cell survives, which simplifies the definition of effective inhibitors.
Researchers have already proved that the peptide inhibitor has penetrated into cancer cells and effectively blocked CJUN. The next step will be pre -clinical trials in cancer models to verify the effectiveness and security of a new approach.
